Description

The inflammasome is a large multiprotein complex whose assembly leads to the activation of caspase-1, which promotes the maturation of proinflammatory cytokines interleukin-1β (IL-1β) and IL-18. The Tribo™ CaspaseFamily fluorometric Assay Kits provides a simple and convenient means for assaying the activity of caspases family members (caspase-1, 2, 3, 4, 5, 6, 8, 9 &10). The assay is based on detection of the free AFC fluorescence after cleavage from the labeled substrate. The free AFC can be quantified using a fluorometer or a fluorescence microtiter plate reader at Ex400/Em505nm. Comparison of the fluorescence of AFC from an apoptotic sample with an uninduced control allows determination of the fold increase in caspase-family member activity.
