Description
Apoptosis plays a fundamental role in many normal biological processes as well as in several disease states. Apoptosis can be induced by various stimuli that all produce the same end result: systematic and orderly cell death. The inflammasome is a large multiprotein complex whose assembly leads to the activation of caspase-1, which promotes the maturation of proinflammatory cytokines interleukin-1β (IL-1β) and IL-18. The Tribo™ CaspaseFamily fluorometric Assay Kits provide a simple and convenient means for assaying the activity of caspases family members (caspase-1, 2, 3, 4, 5, 6, 8, 9 &10). The assay is based on the detection of the free AFC fluorescence after cleavage from the labeled substrate. The free AFC can be quantified using a fluorometer or a fluorescence microtiter plate reader at Ex400/Em505nm. Comparison of the fluorescence of AFC from an apoptotic sample with an uninduced control allows the determination of the fold increase in caspase-family member activity.
Storage Conditions
Store Cell Lysis buffer and Assay buffer at 4℃, Store all other components at -20℃. Shelf life of six months.
