Functions

GRL-0617 is a noncovalent inhibitor of the protease PLpro (papain-like protease; Nsp3) encoded by coronaviruses SARS-CoV (EC50 = 15 µM in Vero cells) and COVID-19 SARS-CoV-2 (EC50 = 28 µM in Vero cells), with no cytotoxicity. Although it was identified in a screen using a fluorescent peptide substrate comprising the 5 C-terminal residues of ubiquitin, it does not inhibit human deubiquitinating enzymes HAUSP, USP18, UCH-L1, UCH-L3, or PLP2.

 

Application Procedures

First dissolved in DMSO (up to 30 mg/ml) or Ethanol (up to 15 mg/ml), then diluted to aqueous buffer. Solutions in DMSO or ethanol may be stored at -20° for up to 3 months.
