Botulinum neurotoxin type A (BoNT-A) is one of the seven serotypes of botulinum neurotoxins (BoNTs) produced by Clostridium botulinum. As zinc proteases, BoNTs cleave SNARE proteins, blocking acetylcholine release at the neuromuscular junction, which can cause flaccid paralysis or even death. The principle of the assay is based on a quantitative sandwich enzyme immunoassay. A monoclonal antibody specific to BoNT-A is pre-coated onto the microplate, binding to target toxin in standards and samples. A biotin-conjugated detection antibody and streptavidin-HRP complex enable signal amplification, followed by color development using an ultra-sensitive TMB substrate. The color intensity, measured at 450 nm, is directly proportional to the amount of BoNT-A present.
