The Tribioscience ATP Colorimetric/Fluorometric Assay Kit (TBS2010) offers a sensitive and straightforward approach to measuring ATP levels in various samples. By utilizing the phosphorylation of glycerol, the kit produces a detectable product that can be measured either colorimetrically at 570 nm or fluorometrically with excitation/emission at 535/590 nm. This dual-detection capability allows for flexibility depending on the sensitivity requirements of the experiment, with the fluorometric method offering 10- to 100-fold higher sensitivity compared to the colorimetric method.

To ensure precise quantification, the kit includes a detailed protocol for preparing standard curves and samples. For instance, standard curves are generated by diluting the provided ATP standard solution to create a range of known concentrations. Samples, such as tissues or cells, are lysed in the assay buffer, and it’s recommended to quick-freeze samples using liquid nitrogen or dry ice if they are to be assayed later, preserving ATP integrity. After lysis and centrifugation to remove insoluble materials, the supernatant is used for the assay. The reaction mix, comprising assay buffer, substrate, probe, and enzyme, is then added to both standards and samples, followed by incubation and measurement.

The kit’s design emphasizes both sensitivity and throughput. With a detection range of 0.5 to 1000 µM in a 96-well plate format, it accommodates a wide variety of sample types and concentrations. The procedure is streamlined to take less than 30 minutes, facilitating high-throughput analyses without compromising accuracy. Proper storage and handling, such as keeping the kit at -20°C and warming components to room temperature before use, further ensure the reliability and reproducibility of results.
