SMO antagonist

Functions

Smoothened (SMO) antagonist that decreased cell proliferation and induced apoptosis (EC₅₀=10 µM) via the intrinsic pathway in esophageal adenocarcinoma cell lines. Ameloblastoma cells with an activating SMO-L412F mutation were insensitive to vismodegib (another SMO antagonist) but BMS-833923 significantly reduced downstream Hedgehog signaling and tumor cell viability. In developmental studies BMS-833923 has been characterized as a uniquely effective SMO inhibitor when it was used to show that Sonic hedgehog is functionally dedicated to ray branching during fin regeneration in zebrafish. Orally active.

Application Procedures

First dissolved in DMSO (up to 25 mg/ml), then diluted to aqueous buffer. Solutions in DMSO may be stored at -20°C for up to 3 months.
