Water-soluble XTT
Water-soluble XTT offers a significant advantage in cell viability assays by eliminating the need for additional solubilization steps, streamlining the workflow for researchers. TribioScience’s XTT Cell Viability Assay Kit utilizes a specially formulated XTT reagent that dissolves readily in aqueous solutions, forming a clear working solution when mixed with an activator at a simple 1:50 ratio. Upon contact with metabolically active cells, the yellow XTT salt is reduced to a bright orange formazan dye through mitochondrial oxidoreductase activity, a process exclusive to viable cells. Since this formazan is water-soluble, absorbance can be measured directly in the assay plate at 450 nm, simplifying analysis and minimizing handling errors.
This water-soluble XTT formulation enhances experimental accuracy and throughput by removing traditionally required wash or cell transfer steps. The entire assay can be conducted in a single 96-well plate, where researchers add the XTT working solution directly to cultured cells and incubate for just 2–4 hours. Because the dye remains in solution, there’s no need for organic solvents or detergents to solubilize the formazan, reducing the risk of variability and contamination.
