Inoue Transformation Buffer for E. Coli component cell
The Inoue Transformation Buffer is a carefully formulated chemical solution whose composition is optimized for the Inoue method. A typical recipe includes MnCl2, CaCl2, KCl, and PIPES. The buffer is stored at 4°C before use; this environment helps stabilize the bacterial cell membranes and primes them to become competent for DNA uptake.
When used in the Inoue method, this buffer enables preparation of competent cells. Cells harvested (after growth at a lower temperature than standard) are washed and resuspended gently in ice-cold Inoue buffer, then treated with a small amount of DMSO, and finally aliquoted and frozen for storage. Under standard conditions, transformation efficiencies in the high-efficiency range are regularly achieved.
Because of this optimized buffer-mediated preparation, the Inoue method offers a balance of high efficiency, reproducibility, and convenience: labs can prepare their own competent cells rather than relying on commercial ones, with transformation yields sufficient for most cloning tasks.
