This is crucial after aminopterin in the HAT medium has inhibited the de novo nucleotide synthesis pathway, ensuring only hybrid cells with functional salvage pathways survive and thrive.

The Hybridoma Growth Medium with HT exemplifies such a specialized formulation. It is a chemically defined, animal component-free liquid medium containing hypoxanthine, thymidine, L-glutamine, gentamicin, and phenol red. For optimal use, the medium should be thawed at room temperature or overnight at 2–8°C, avoiding thawing at 37°C to maintain its integrity.
