Dispase II Powder is a lyophilized enzyme preparation designed for convenient storage, rapid reconstitution, and reliable performance in tissue dissociation applications. The freeze-dried (lyophilized) format ensures long-term stability and easy handling, allowing researchers to prepare fresh solutions as needed without compromising enzyme activity. Its consistent performance makes it ideal for generating viable single-cell suspensions from various tissue types.

Reconstitution of Dispase II Powder is simple and efficient. To prepare a 10 mg/mL stock solution, dissolve the nonsterile lyophilized enzyme in Dulbecco’s Phosphate Buffered Saline (DPBS). The reconstituted stock solution remains stable for up to two weeks when stored at +2 to +8°C. For longer-term storage, up to two months, the stock can be frozen in aliquots, minimizing freeze-thaw cycles and preserving enzymatic activity.

To create a working solution, dilute the 10 mg/mL stock with DPBS or the culture medium intended for the target cells, achieving a final concentration between 0.6 and 2.4 U/mL. Concentrations above 2.4 U/mL are not recommended to maintain optimal cell viability. For best results, filter the working solution through a 0.22 μm pore-size membrane to ensure sterility and consistency in performance.
