Beta-NAG High-throughput screening
The β-N-Acetylglucosaminidase (β-NAG) Activity Fluorometric Assay is specifically designed for high-throughput screening (HTS), enabling rapid and reliable quantification of β-NAG activity across thousands of biological samples daily. This lysosomal enzyme, expressed in organs such as the kidneys, liver, lungs, and mast cells, plays a key role in several physiological and pathological conditions, including Tay-Sachs and Sandhoff diseases, as well as immune and inflammatory disorders. The assay leverages a sensitive fluorescence-based detection method using the non-fluorescent substrate 4-Methylumbelliferyl N-acetyl-β-D-glucosaminide (4-MUAG), which, upon enzymatic cleavage, releases the highly fluorescent 4-Methylumbelliferone (4-MU). Fluorescence is then quantified at an excitation of 365 nm and an emission of 445 nm.
Tailored for high-throughput workflows, this assay supports seamless integration with automated liquid handling systems. It is optimized for speed and sensitivity, delivering a linear detection range within a 30-minute reaction at 37°C using just 20 µL of sample.
