Hypoxanthine-Thymidine Medium
Hypoxanthine-Thymidine (HT) Medium is a selective, chemically defined culture medium designed to support the growth and expansion of hybridoma cells following HAT (hypoxanthine-aminopterin-thymidine) selection. This medium contains two essential components—hypoxanthine and thymidine—that are crucial for sustaining hybridoma cell proliferation once aminopterin, the agent used during the selection process, is removed. By omitting aminopterin and continuing supplementation with HT, the medium ensures a smooth transition for hybridomas, promoting stable expansion and viability.
The Hybridoma Growth Medium with HT is animal-derived component-free and optimized for high cell yield and monoclonal antibody (MAb) productivity. In addition to hypoxanthine and thymidine, it includes gentamicin for antimicrobial protection, phenol red as a pH indicator, L-glutamine for metabolic support, and other defined ingredients. This formulation ensures consistency and reproducibility across batches, which is especially important in large-scale or regulated bioproduction environments. Its design specifically addresses the cellular needs of hybridomas post-selection, making it ideal for laboratories focused on antibody generation and hybridoma research.
This medium is supplied in 500 mL units and should be stored at -20°C for up to one year. For use, it can be thawed at room temperature or overnight at 2–8°C, avoiding high-temperature thawing methods. Once thawed, the medium can be stored at 2–8°C for up to two weeks or re-aliquoted and frozen again for extended use.
