High-sensitivity ADP assay

Our high-sensitivity ADP assay is engineered to detect even trace amounts of adenosine diphosphate (ADP) with minimal background noise, ensuring precise and reliable measurements. Utilizing a robust enzymatic reaction, the assay converts ADP to ATP and pyruvate, with the resulting pyruvate quantified either colorimetrically at 570 nm or fluorometrically at Ex/Em 530/590 nm. This dual-detection capability allows for flexibility in experimental design and sensitivity requirements. Notably, the fluorometric method offers 10- to 100-fold higher sensitivity compared to the colorimetric approach, enabling detection of ADP concentrations as low as 1 µM in various biological samples.

Designed for simplicity and efficiency, the assay requires only 10 µL of sample per well and can be completed in under 60 minutes. Its straightforward “load and incubate” protocol eliminates the need for complex preparation steps, making it highly suitable for high-throughput screening applications. The assay’s stability and adaptability ensure consistent performance across a range of experimental conditions, providing researchers with a dependable tool for monitoring ADP levels in cells and other biological samples.

The kit includes all necessary components for 100 tests, such as the ADP assay buffer, probe, substrate, enzyme mix, and a 50 mM ADP standard. For optimal performance, it is recommended to store the kit at -20°C, with a shelf life of three months. Prior to use, all components (except the enzyme) should be brought to room temperature, and small vials should be briefly centrifuged.
